      Time required
        - 4~5 weeks after sample condition check.
 
Step 1 : DNA preparation
  Separates high molecular weight DNA (100~400kb) from the host.
   
Step 2 : Insert preparation 1
Random shearing: Secures the insert with the desired size through size selection in agarose gel after random shearing using a nebulizer.
       
Step 3 : Insert preparation 2
  Processes the insert to be ligated to the vector appropriately.
   
Step 4 : Ligation
  After ligation to cosmid vector, packages the recombinant cosmid DNA into bacteriophage lamda particles using Lamda packaging extract in vitro and constructs a library by transducing them to an appropriate host.
