Step 1 : DNA preparation

Genomic shotgun library: Separates high molecular weight DNA (100~400kb) from the host.
Large costruct shotgun library (BAC, Cosmid, Fosmid, Plasmid, etc.): Separates the sample DNA from the host and removes the host genomic DNA. Checks the samples based on the size of the prepared sample DNA and the results of end sequencing.

   
Step 2 : Insert preparation 1

Random shearing method: Secures the insert with the desired size by size-selecting DNA(2~5kb) obtained through hydroshear random shearing using agarose gel electrophoresis once again
       
Step 3 : Insert preparation 2
  Processes the insert to be ligated to the vector appropriately (blunting or CIP process).
   
Step 4 : Ligation
  Checks the quality of the library by ligating it to high copy vector or low copy vector.
